Pleural mesothelioma is a rare disease. For 17 years platinum-based chemotherapy has been the standard for the treatment of unresectable or metastatic pleural mesothelioma. With the results of the randomized Checkmate 743 phase 3 trial, a new palliative treatment option with checkpoint inhibition has been approved in the first-line setting. Current clinical trials are evaluating combination therapies with checkpoint inhibition in the first line setting or in other settings.

Kurzfassung: Das Pleuramesotheliom zählt zu den seltenen Krebserkrankungen. Für die letzten 17 Jahre war der Therapiestandard für nicht resektable oder metastasierte Stadien eine platinhältige Chemotherapie in Kombination mit Pemetrexed. Mit den Ergebnissen der randomisierten Phase-3-Checkmate-743-Studie steht nun eine neue palliative Therapieoption mittels dualer Checkpoint-Blockade in der Erstlinie zur Verfügung. Aktuelle Studien untersuchen Kombinationstherapien mit Checkpoint-Inhibitoren in der Erstlinie bzw. auch in anderen Behandlungslinien.
