Präimplantationsdiagnostik (PID) für chromosomale Aberrationen und monogene Erkrankungen – Ergebnisse eines deutschen PID-Zentrums // PGT for chromosomal aberrations and monogenic disorders – results of a German PGT center Journal für Reproduktionsmedizin und Endokrinologie - Journal of Reproductive Medicine and Endocrinology 2026; 23 (5): 191-199 Volltext (HTML) Summary Abbildungen Keywords: Aneuploidie, monogene Erkrankung, Präimplantationsdiagnostik, reziproke Translokation, Robertson’sche Translokation, aneuploidy, monogenic disorder, Preimplantation Genetic Testing, reciprocal translocation, Robertsonian translocation Preimplantation Genetic Testing (PGT) is the genetic examination of trophoblast cells (trophectoderm, TE) of day 5 or day 6 blastocysts. PGT is legally regulated by the embryo protection act (ESchG) [1], which allows genetic testing for specific indications of embryonic cells at state-approved PGT centers after approval of PGT ethics committee. It enables the genetic proof of chromosomal aberrations and pathogenic gene variants of an embryo and thus prevents from pregnancies with an affected child. The present study represents data of the PGT cases carried out at the Medical Genetics Center over the period from July 2015 to December 2025. It focusses on PGT for structural chromosomal aberrations of couples with balanced reciprocal and Robertsonian translocations (preimplantation genetic testing for structural rearrangements, PGT-SR). It could be shown that the percentage of abnormal and normal results in trophectoderm samples (TE samples) differ depending on wether it is a reciprocal or a Robertsonian translocation and wether it is a maternal or paternal translocation. This study also represents results of PGT for numerical chromosome aberrations (preimplantation genetic testing for aneuploidies, PGT-A, and PGT for monogenic disorders, PGT-M). It emphasises the need of concomitant testing for pathogenic gene variants and chromosomal aberrations. Although the percentage of abnormal results in TE samples was high, at least one embryo could genetically be recommended for transfer in more than two thirds of all cycles, as in the vast majority of cycles at least three blastocysts have been analysed on average. Preimplantation genetic testing significantly reduces the risk of passing on severe hereditary conditions by screening embryos before implantation.
